Factor 1 — Importance of the Research
scored 1–9- Reviewers are asked
- Is there a strong scientific premise, and would successful completion change the field? (This is the old Significance + Innovation, merged.)
- So write
- State the problem, the gap, and what specifically becomes possible if you succeed. Cite the evidence your premise rests on — and be explicit where prior work is weak.
- Commonly loses points for
- Premise rests on a single unreplicated finding, or on the applicant's own unpublished data only
- "Innovation" is a new technique applied to a question nobody was blocked on
- Significance is asserted ("this is critically important") rather than demonstrated
- Incremental extension of the applicant's prior R01 with no new conceptual advance
Factor 2 — Rigor and Feasibility
scored 1–9- Reviewers are asked
- Is the approach rigorous, and can this team actually do it in the time and budget? (The old Approach criterion.)
- So write
- Per aim: hypothesis, design, sample sizes with power justification, controls, analysis plan, expected outcomes, and what you do if the result is negative. Include preliminary data that proves the hardest step works.
- Commonly loses points for
- No alternative strategy — reviewers read a single-path plan as high risk
- Missing power analysis or biological replicates; n chosen for convenience
- Aim 2 and Aim 3 depend entirely on Aim 1 succeeding (linear dependency)
- Overambitious scope for the budget period — the single most common critique
- Rigor boilerplate (sex as a biological variable, blinding, randomisation) omitted
Factor 3 — Expertise and Resources
sufficient / not sufficient- Reviewers are asked
- Are the team and environment sufficient for THIS project? Reviewers mark sufficient or not sufficient and must explain if not — deliberately no numeric score, to reduce the pull of reputation.
- So write
- Map each required capability to a specific person or facility. If you lack a skill, name the collaborator who has it and include their letter.
- Commonly loses points for
- A required method (e.g. cryo-ET, subtomogram averaging) has no one on the team who has published it
- Instrument access asserted without a facility letter or documented allocation
- Collaborator listed with no letter confirming their role
Section-by-section outline
1.Specific Aims
1 pageThe only page every reviewer reads carefully. It must stand alone as the whole argument.
Must include
- Opening paragraph: the problem and why it matters now
- Gap paragraph: what is unknown, and why existing approaches have not closed it
- Your central hypothesis and the objective of this application
- 2–3 aims, each with a one-line rationale, approach, and expected outcome
- Closing payoff: what the field can do after this work that it cannot do today
Avoid
- Aims that are methods ("we will perform cryo-EM") rather than questions
- Interdependent aims — if Aim 1 fails, the application fails
- No stated hypothesis, only a list of activities
2.Significance
~1 page of Research StrategyEstablish scientific premise and the consequence of success (feeds Factor 1).
Must include
- The specific knowledge gap, with citations
- A candid assessment of the strength of the prior literature
- What downstream work your result unblocks
Avoid
- Reviewing the field instead of arguing for this project
3.Innovation
~0.5 pageSay what is genuinely new — concept, method, or both (feeds Factor 1).
Must include
- What is novel
- Why now (what changed technically to make this possible)
Avoid
- Claiming novelty for a technique that is standard in another field
4.Approach
~6–8 pages (12-page Research Strategy for R01)The bulk of the score sits here (Factor 2).
Must include
- Preliminary data establishing feasibility of the hardest step
- Per aim: design, controls, sample size + power, statistics, expected results
- Potential problems and alternative approaches for each aim
- Timeline and milestones
Avoid
- No alternatives section
- Statistics deferred to "standard methods will be used"
- Scope that cannot be delivered in the project period
Strategy notes for this mechanism
- Factor 3 being unscored means a strong CV no longer lifts a weak Approach — put your effort into Factor 2.
- Contact the programme officer before writing. Fit with the institute's priorities is decided outside the score.
- For resubmissions, the Introduction responding to critiques is read first — address every point explicitly.
